HIV antigens are immobilized on a permeable immunofiltration layer. Test and reagents go through the film and are absorbed into the underlying absorbent. As the patients sample goes through the layer HIV antibodies, if present bind to the immobilized antigens. Conjugate ties to the Fc portion of the HIV antibodies to give particular pinkish purple DOT(s) against a white foundation. Other related techniques include western blot test, quantitative dot blot, immunohistochemistry and immunocytochemistry where antibodies are utilized to identify proteins in tissues and cells by immune staining, and Enzyme-Linked Immunosorbent Assay (ELISA)
